Due to their high affinity, specificity, and relatively cheap and fast synthesis, aptamers and aptamer-based systems have been developed in recent years for numerous analytical and medical applications. After an introduction and common protocols for aptamer synthesis, different optimisation strategies to obtain higher affinities to target molecules are presented. Characterization of aptamer-target molecule interaction and immobilization strategies are the foundation of an in-depth discussion of method developments for all relevant applications, from colorimetric methods to lateral flow immunoassays.
